Buying Nitrosamine Reference Standards: What to Ask First

Nitrosamine standards are small in quantity but big in consequence. The questions to put to any supplier before an NDMA or NDSRI standard reaches your lab.

Nitrosamine testing has become routine in pharmaceutical quality control, but buying the standards still causes delays. Many nitrosamine drug substance related impurities, usually called NDSRIs, are not available off the shelf. Others are offered by several suppliers with very different levels of supporting data. A few milligrams of the wrong material can cost weeks of method development.

This article is not about the regulatory limits themselves. It focuses on the purchasing step: what to ask a supplier, what to look for in the documents and how to plan around lead times.

Start by defining exactly what you need

Before you contact anyone, pin down the structure. For small, well-known nitrosamines such as NDMA, NDEA, NDIPA, NEIPA, NMBA or NDBA, the name and CAS number are usually enough. For an NDSRI, send a structure drawing or SMILES string as well as the name, because these compounds often have no CAS number yet, and different suppliers name them differently.

Also decide which form you want:

  • Neat material in milligram quantities, which gives you full control over solution preparation.
  • A certified solution, usually in methanol, which reduces handling of a potent compound but limits your choice of diluent.
  • A stable-isotope labelled analogue, such as NDMA-d6, to use as an internal standard in LC-MS/MS or GC-MS methods.

Questions about identity

The identity of a nitrosamine standard must be established beyond reasonable doubt, because the whole risk assessment depends on it. Ask the supplier:

  1. Which techniques confirmed the structure? Expect at least 1H NMR and mass spectrometry, and often 13C NMR and IR.
  2. Was the position of the N-nitroso group confirmed? For NDSRIs derived from molecules with more than one secondary amine, nitrosation can occur at different sites, giving different isomers.
  3. Are spectra included with the certificate, or only summarised as "conforms"?

The rotamer question

Asymmetric nitrosamines exist as E and Z rotamers because of restricted rotation around the N-N bond. In NMR this produces doubled signals, and in some chromatographic conditions it can give split or broadened peaks. A good supplier will explain this in the data package. If they do not, and you see two peaks, you may waste time investigating a non-existent impurity. Ask whether rotamers are observed and how the purity figure accounts for them.

Questions about purity and content

  • How was purity assigned? Chromatographic purity alone is not a content value. For quantitative work you want a figure that also accounts for water, residual solvents and inorganic content, or a quantitative NMR result.
  • Is the value on an as-is basis? You will use the figure in calculations, so its basis must be clear.
  • What is the uncertainty? For trace methods near the acceptable intake limit, the uncertainty of the standard contributes directly to your result.

Questions about stability and handling

N-nitroso compounds can be sensitive to ultraviolet light, which breaks the N-N bond. Some NDSRIs are also thermally fragile or prone to denitrosation in acidic solution. Ask for:

  • Recommended storage temperature and whether protection from light is required.
  • Any stability data or retest date for the neat material.
  • Guidance on solution stability in common diluents.

Small volatile nitrosamines such as NDMA also evaporate easily from open vials. Weigh them quickly, or buy a certified solution, and keep stock solutions tightly closed and cold.

Finally, ask whether the supplier can provide the same lot again later. Methods are often transferred between sites or revalidated years after development, and a consistent lot avoids having to re-establish relative response factors or retention behaviour.

Questions about safety and shipping

Many nitrosamines are classified as probable human carcinogens. Expect a full GHS safety data sheet, and plan for handling in a fume hood or isolator. Milligram quantities usually travel as non-dangerous or excepted quantities by courier, but solutions in flammable solvents may carry transport restrictions. Confirm the shipping method before ordering if you are on a tight timeline.

Planning around lead times

Catalogue nitrosamines can usually be supplied quickly. Custom NDSRIs are different. A typical route involves synthesising or sourcing the parent amine, nitrosation, purification and full characterisation. Even when the chemistry is straightforward, the analytical work takes time. Practical tips:

  1. Order the standard as soon as the risk assessment flags a possible NDSRI, not when the method is due.
  2. Request a labelled analogue at the same time if your method will need one, since it often requires a separate synthesis.
  3. Agree the data package in writing before synthesis starts.
  4. Ask for enough material for method validation and future system suitability, not just development.

Checking the material when it arrives

Even with a strong certificate, a short receiving check is worthwhile for standards that will support regulatory submissions.

  • Confirm that the structure, name and lot on the label match the certificate and your order.
  • Run a quick chromatographic check in your own method. Retention time, peak shape and any rotamer behaviour should match the supplier's description.
  • If you use LC-MS/MS, confirm the expected precursor and product ions and record them in the method file.
  • Store the material immediately at the stated temperature, protected from light, and log the receipt date.

Keep the full supplier data package with your method validation records. Inspectors reviewing a nitrosamine risk assessment often ask how the identity of the standard was established, and having the spectra to hand avoids a scramble.

Where Zemixal fits

Zemixal lists a wide range of nitrosamine and other impurity reference standards, supplied with characterisation data appropriate to the product, typically HPLC, NMR, MS and IR. Browse the impurity standards section or search by parent drug name. For NDSRIs that are not listed, send the structure through the RFQ form and ask about custom synthesis. If you are screening many candidate impurities at once, paste their CAS numbers into the bulk CAS lookup to see which are already in the catalogue.

Questions readers ask

Why do NDSRIs often lack a CAS number?
Many were first described recently, during risk assessments for specific drugs, and have not yet been registered. Suppliers therefore identify them by structure, name and an internal catalogue number.
Is a deuterated nitrosamine always the best internal standard?
It is the usual choice, but check that the label sits on non-exchangeable carbon positions and that the unlabelled content is low enough not to bias trace results. 13C or 15N labels avoid exchange entirely but cost more.
How much NDSRI standard should we order?
Enough for development, validation and at least a year of routine system suitability. Ten to twenty-five milligrams is common for neat material, but your method concentration and number of sites decide the real figure.
