Pain and inflammation products are among the highest-volume generics, and their APIs need consistent physical properties as well as chemical purity. Why is particle size specified for NSAID APIs?
Many NSAIDs are poorly soluble in water, so their dissolution rate depends heavily on particle size. Smaller particles dissolve faster and can raise bioavailability, but may flow poorly during tableting. Formulators therefore set a particle size distribution, often by laser diffraction, as part of the API specification.
What is special about dexketoprofen compared with ketoprofen?
Dexketoprofen is the active S-enantiomer of ketoprofen, usually supplied as the trometamol salt. Using the single enantiomer delivers the analgesic effect at a lower dose. Its specification therefore includes enantiomeric purity, measured by chiral HPLC, in addition to the usual assay and impurity tests.
