Pyridine is one of the most common rings in approved drugs, valued for its basic nitrogen, hydrogen bond acceptance and metabolic stability. This sub-category supplies pyridines with a wide range of substitution patterns and functional handles. Examples include (5-Bromo-3-methoxypyridin-2-yl)methanol, (6-(Aminomethyl)pyridin-2-yl)methanol dihydrochloride, 1-(4-Pyridyl)piperazine, 1-(3-Hydroxypyridin-4-yl)ethanone and the pyridinone 1-(4-Methoxyphenyl)-5-methyl-2(1H)-pyridinone.

Substitution pattern is everything with pyridines. The 2-, 3- and 4-positions differ sharply in reactivity, so halogens at C-2 or C-4 undergo substitution easily, while those at C-3 need cross-coupling. Buyers should check the exact positions against the drawing, as names can be confusing. Many aminopyridines are supplied as hydrochloride salts for stability, so adjust weights for the counter-ion when planning reactions.

Products ship in sealed bottles from milligram to kilogram scale. Each lot comes with a COA and SDS. For larger libraries, paste your CAS list into the Bulk CAS lookup to check availability in one go.

Pyridine Derivatives: questions buyers ask

Why are 2-halopyridines more reactive than 3-halopyridines?
The ring nitrogen withdraws electron density from the 2- and 4-positions and stabilises the negative intermediate formed during nucleophilic substitution. Halogens at those positions are displaced by amines and alkoxides under mild conditions. A halogen at C-3 lacks this activation and normally needs palladium or copper catalysis.
How do I free-base an aminomethylpyridine dihydrochloride?
Dissolve the salt in water, add a base such as sodium hydroxide or carbonate until strongly basic, and extract the free amine into an organic solvent such as dichloromethane. Dry and concentrate carefully. Alternatively, add a tertiary amine base directly to the reaction to release the amine in situ.
Are pyridinones the same as hydroxypyridines?
2- and 4-hydroxypyridines exist mainly in their pyridinone tautomer form, with a carbonyl and an N-H or N-substituent. N-substituted pyridinones, such as the methoxyphenyl example, are locked in that form. This affects reactivity, hydrogen bonding and solubility, and pyridinones appear in drugs such as pirfenidone.
