Six-membered rings with two nitrogens form the core of nucleic acid bases and many modern drugs. This sub-category covers pyrimidines, pyrazines and pyridazines along with fused systems. Examples include 2,4,6-Trichloropyrimidine, 2,4-Diamino-6-hydroxypyrimidine, 1,3-Dimethyluracil, 1H-Pyrazolo[3,4-d]pyrimidin-4-amine (a kinase scaffold), 2'-Deoxy-5-fluorouridine and 2,3-Pyrazinedicarboxylic acid.

Chloropyrimidines are especially useful because each chlorine reacts at a different rate, allowing stepwise substitution with amines, alcohols and thiols. The 4-position usually reacts first, then the 2-position, under controlled conditions. Aminopyrimidines and uracils, by contrast, are used for condensation and nucleoside chemistry. Purity and absence of over-chlorinated or hydrolysed impurities are the main specification points, and both are reported on the lot COA.

  • Polychloropyrimidines for sequential substitution
  • Pyrazolopyrimidines and aminopyrimidines as kinase scaffolds
  • Uracils and nucleosides for antiviral and oncology research

Each lot ships with a COA and SDS. Chloropyrimidines are moisture sensitive and are packed dry, and kilogram quantities can be quoted for process work.

Pyrimidines, Pyrazines & Pyridazines: questions buyers ask

In which order do the chlorines of 2,4,6-trichloropyrimidine react?
The 4- and 6-positions are the most reactive towards nucleophiles and react first, usually at low temperature. The 2-position reacts later under more forcing conditions. Careful control of temperature and stoichiometry lets you introduce three different groups in sequence, a common strategy in kinase inhibitor synthesis.
What is 1H-pyrazolo[3,4-d]pyrimidin-4-amine used for?
It is an adenine analogue and a core scaffold for kinase inhibitors, including the BTK inhibitor ibrutinib. The scaffold mimics ATP binding in the kinase hinge region. Chemists substitute it at the 1- and 3-positions to tune potency and selectivity against different kinases.
How should chloropyrimidines be stored?
Keep them tightly sealed, dry and cool, preferably under nitrogen. Moisture slowly hydrolyses the chlorines to hydroxyl groups, giving pyrimidinones that interfere with substitution. Many chloropyrimidines are also skin sensitisers and irritants, so handle them with gloves in a fume cupboard and follow the SDS.
