Five-membered nitrogen rings appear in proline, many alkaloids and drugs ranging from DPP-4 inhibitors to anti-inflammatories. This sub-category covers aromatic pyrroles and pyrazoles alongside saturated pyrrolidines and pyrrolidones. Examples include (R)-(+)-α,α-Diphenyl-2-pyrrolidinemethanol, a CBS catalyst precursor, (S)-1-(2-Aminoacetyl)-4,4-difluoropyrrolidine-2-carbonitrile hydrochloride, (S)-Ethyl-N-Boc-pyroglutamate, 1-(Difluoromethyl)-4-nitro-1H-pyrazole and 1-(Benzenesulfonyl)pyrrole.

Chiral pyrrolidines usually derive from proline, so their configuration and ee should be confirmed on the COA. Pyrazoles bring two adjacent nitrogens and are often substituted with fluorinated groups to tune drug properties. N-protected pyrroles such as the benzenesulphonyl derivative allow selective substitution at the 3-position that would otherwise be hard to reach. Pyrrolidones such as N-butyl pyrrolidone also serve as polar aprotic solvents with a lower hazard profile than NMP.

  • Prolinol derivatives for asymmetric catalysis
  • Cyanopyrrolidines for DPP-4 inhibitor research
  • Fluorinated pyrazoles for drugs and agrochemicals

Each lot ships with a COA and SDS, sealed and cold-packed where needed.

Pyrroles, Pyrrolidines & Pyrazoles: questions buyers ask

What is diphenylprolinol used for?
α,α-Diphenyl-2-pyrrolidinemethanol reacts with borane sources to form CBS oxazaborolidine catalysts for enantioselective ketone reduction. Its silyl ethers are also very widely used organocatalysts for asymmetric Michael additions and alpha-functionalisation of aldehydes. The R and S forms give opposite product configurations.
Why are cyanopyrrolidines important in diabetes drugs?
The 2-cyanopyrrolidine unit binds the active site of the enzyme DPP-4 and is found in gliptins such as vildagliptin and saxagliptin. Difluoro substitution on the ring can improve potency and stability. Researchers use these building blocks to make analogues and impurity standards of these drugs.
Why protect the pyrrole nitrogen with a sulphonyl group?
Free pyrrole reacts mainly at the 2-position and can polymerise under acid. A benzenesulphonyl group withdraws electron density, stabilises the ring and steers electrophilic substitution towards the 3-position. The protecting group is removed later with base, revealing the free N-H pyrrole.
